Dear All,
We at TIGR are in the process of constructing new Tetrahymena thermophila cDNA libraries for the
purpose of EST sequencing. The more EST sequences we have, the better we can train gene finder
programs to improve the annotation of the genome sequence, which will benefit the entire
community. In order to cover as many genes as possible, we are constructing libraries from cells
in different developmental or environmental conditions. Currently, we are working on RNA from
cells that were conjugating, starved, and stressed (by exposure to toxins and DNA-damaging
agents). If you have additional suggestions of treatments that you expect would induce novel sets
of genes (and especially if you would be willing to provide whole cell RNA for the construction of a
library) please let me know. In particular, we are interested in constructing a library from cells
grown in minimal medium (recipe available in the Tetrahymena Methods in Cell Biology, Vol. 62
book). If anyone is using this or a similar medium, or has used one in the recent past, please
contact me.
